fa بررسی تمایز و ساختار ژنتیکی ماهی سوف معمولی (Sander lucioperca) در دریاچه سد ارس و تالاب انزلی تخصصي Special پژوهشي Research هدف این مطالعه بررسی تمایز و ساختار ژنتیکی جمعیت ماهی سوف معمولی (Sander lucioperca) در دو منطقه تالاب انزلی و دریاچه سد ارس بود. در کل 60 نمونه ماهی سوف معمولی بالغ از این دو منطقه در تابستان 91 جمع آوری شدند. از پنج جفت پرایمر میکروستلایت، بر روی DNA ژنومی ماهی سوف معمولی استفاده گردید که همگی پرایمرها چند شکل (پلی‌مورف) نشان دادند و از آن‌ها برای تعیین تمایز ژنتیکی استفاده شد. میانگین اللی (Na) در جایگاه‌ها 5 (با دامنه 4 تا 7 الل) بود. هر دو منطقه نمونه‌برداری اللهای اختصاصی در تمامی جایگاه‌ها نشان دادند. میانگین هتروزیگوسیتی قابل انتظار و مشاهده شده به ترتیب 731/0 و 650/0 محاسبه شد. میانگین ضریب خویشاوندی در 5 جایگاه میکروستلایت منفی بود. در بررسی تعادل هاردی وینبرگ (H-W) تمامی جایگاه‌ها به‌طور معنی‌داری خارج از تعادل هاردی-وینبرگ بودند (001/0≥P). میزان شاخص تمایز (Fst) و جریان ژنی (Nm) بر اساس فراوانی اللی به ترتیب 092/0 و 4/2 محاسبه شد. بر اساس آزمون AMOVA میزان Rst و Fst بین جمعیت‌های دو منطقه معنی‌دار بود (01/0≥P). میانگین ضریب خویشاوندی (Fis) در پنج جایگاه منفی بود. میزان فاصله ژنتیکی بین جمعیت‌ها 616/0 به دست آمد که نشان دهنده تمایز ژنتیکی بین جمعیت‌های مورد مطالعه است. این بررسی، وجود تنوع و تمایز ژنتیکی جمعیت های ماهی سوف معمولی در تالاب انزلی و دریاچه سد ارس را نشان داد. This study aimed to determine the population genetic variation of pikeperch, Sander lucioperca, investigated in Anzali Wetland and Lake of Aras dame using microsatellite markers. Totally 60 samples of adult pikeperch were collected from these regions in summer 2012. Five sets of microsatellite primers tested on genomic DNA of pikeperch. All primer sets as polymorphic loci were used to analyze the genetic variation. Analyses revealed that average of alleles (Na) per locus was 5 (range 4 to 7 alleles). All sampled regions contained private alleles. The average observed and expected heterozygosity was 0.650 and 0.731 respectively. The average estimates of inbreeding coefficient (Fis) values of five microsatellites were negative. Deviations from Hardy-Weinberg equilibrium were in all cases (P<0.01). F-statistics (Fst) and gene flow (Nm) estimates in Allele frequencies were 0.092 and 2.4 respectively. Rst and Fst estimates in AMOVA indicated significant genetic differentiation among regions (P≤0.01). Genetic distance was 0.616, indicating that the genetic difference among the studied populations is pronounced.
